Glass ampules are made available with G. stearothermophilus spores that can be used in steam-based processes. The ampules hold spores in a solution of culture broth. Since the spores and the media are mixed, no crushing of the vial or activation is needed after processing. Incubation for 48 hours is recommended. However, it is typical for growth to occur within 8-12 hours. Spore growth can be observed by a yellow-colored change in the medium, along with visible turbidity near the inside of the bottle.

The Biological indicator is then subjected to the sterilization process and is then incubated under specific growth conditions to determine if any spores survive the process. If no spores are found, there is no growth, and the test is considered a pass. If growth is observed, then the test is considered a failure.

How Do Biological Indicators Work for the Sterilization Process?

Biological indicators are typically used for routine monitoring, calibration, and load monitoring for steam sterilizers. They are used to determine whether the conditions present during the steam cycle were sufficient to attain a specified level of microbial inhibition. For sterilization by steam, the majority of biological indicators are utilized in PCDs.

The PCD with the biological indicator is put in the most demanding spot within the chamber. For a steam sterilizer, it is usually placed on the lower shelf, close to the drain. 

Biological indicators must be utilized to ensure the regular monitoring of the steam sterilization cycle once a week at a minimum, but preferably daily, and for every load that has implants.
